Ever leave your amp on overnight?

You might, but I wouldn't think enough to make a big difference.

As long as your amp was in standby, there was no high voltage going to the power tubes, only heater voltage, in other words, your power tubes were idling so to speak.

Back in the day, those old tube powered PA amps ( Bogen, EV, etc. like you used to see in a closet at church or something) ran for years and years, many times never getting turned off, but, tubes where more reliable then too.

yes, thank you. tubes wear for only one reason-heat to the metal. as the metal is hot it causes some of the metal particles to shed, and that is the grey stuff you see gathering in the glass, usually down by the pins. but what causes this to happen the most is the expanding and contracting of the metal by turning it off and on, more so than staying left on. just a rough estimation, but turning your amp on and off say 3 times for a total of an hours worth of play is roughly equal to leaving it on for a full 24 hours, as far as tube wear goes.
