Put your logic hat on, and use your head. Start eliminating things, one by one. If you haven't already done so, just plug the guitar straight into the amp. Try a different cord. Try a different guitar. Try the speaker with a different amp. Try the most obvious of things, such as amp standby switch and speaker connection. Finally, if it's none of those, it's probably the output tube.

And the most obvious one. Have you tried to reseat the tube?

 

One of the reasons I turned all my VJ heads upside down...(besides the better heat dissipation).

First, check the heater fuse. The orange wire has a fuse on it. Open that up, sand the ends a bit, and put it back together. Sometimes, a varnish on the ends of that fuse prevent it from making contact, and it will do what you're describing.

 

The output tube is the taller of the two.
